چکیده
Green fluorescent protein, GFP, is a spontaneously fluorescent protein isolated from coelenterates, such as the Pacific jellyfish, Aequorea victoria, or from the sea pansy, Renilla reniformis. Its role is to transduce the blue chemiluminescence of aequorin, into green fluorescent light by energy transfer. The molecular cloning of GFP cDNA and the expression of GFP as a functional transgene has opened exciting new avenues of investigation in cell, developmental and molecular biology. GFP can function as a protein tag, as it tolerates Nand C-terminal fusion to a broad variety of proteins many of which have been shown to retain native function. Highly specific intracellular localization including the nucleus, mitochondria, secretory pathway, plasma membrane and cytoskeleton can be achieved via fusion both to whole proteins and individual targeting sequences. The enormous flexibility as a non-invasive marker in living cells allows for numerous other applications such as a cell lineage tracer, reporter of gene expression and as a potential measure of protein-protein interactions.

متن کاملمهار بیان ژن GFP به وسیله تداخل RNA (RNAi) در دودمان سلولی کارسینومای جنینی P19
Introduction: RNA interference (RNAi) is a phenomenon of gene silencing that uses double-stranded RNA (dsRNA), specifically inhibits gene expression by degrading mRNA efficiently. The mediators of degradation are 21- to 23-nt small interfering RNAs (siRNA). The use of siRNAs as inhibitors of gene expression has been shown to be an effective way of studying gene function in mammalian cells. Ai...
متن کاملDetection of cancer cells using triplex DNA molecular beacons based on expression of enhanced green fluorescent protein (eGFP).
A novel strategy was proposed for Ramos cell detection by combining the expression of enhanced green fluorescent protein (eGFP) with the cell aptamer recognition and the triplex molecular beacons. This system was successfully applied to cancer cell detection with high sensitivity and specificity.
متن کاملCap analog and Potato virus A HC-Pro silencing suppressor improve GFP transient expression using an infectious virus vector in Nicotiana benthamiana
Transient expression of proteins in plants has become a choice to facilitate recombinant protein production with its fast and easy application. On the other hand, host defensive mechanisms have been reported to reduce the efficiency of transient expression in plants. Hence, this study was designed to evaluate the effect of cap analog and Potato virus A helper component proteinase (PVA HC-Pro) o...
متن کاملTransfection of bovine spermatogonial stem cells in vitro
Spermatogonial stem cells (SSCs) are the only stem cells in adults that can transfer genetic information to the future generations. Considering the fact that a single SSC gives rise to a vast number of spermatozoa, genetic manipulation of these cells is a potential novel technology with feasible application to various animal species. The aim of this study was to evaluate enhanced green fluoresc...
